Experimetria currently offers many whole heart perfusion systems to choose from, which are designed for the in-vitro investigation of the heart of small animals such as rats, hamsters, guinea-pigs, rabbits up to dogs (maximum weight is approximately 3 grams). The first system is a Langendorff based on the Janicki method (as described by Janicki JS), also called the retrograde perfusion method, where the heart is not actually working. The second is based on the Neely working heart method (as described by Neely JR). The Langendorff system has the cannula attached to the aorta and LVP is measured by insertion of a balloon into the left ventricle. The aortic valve remains closed, hence the retrograde perfusion (non-working heart) design. This system can be either a constant pressure or constant flow system or both. The standard Langendorff configurations Experimetria offers are two column designs allowing for ischemia studies (with the second column bubbling nitrogen), or precise drug effect studies, utilizing a quick valve control to switch between the two columns. Lower cost single column options are also available for non ischemic research or drug effect studies using direct drug introduction. The Working Heart system has the cannula attached to the left atrium allowing for proper flow within the heart, hence the working design. LVP is measured with the insertion of a "Millar" type catheter. The Working Heart system is a two column design that can also operate as a Langendorff in either constant pressure or constant flow modes. In fact when the heart is first attached to the Working Heart system it is first perfused in a constant pressure Langendorff mode, before it is actually worked. Installation by an Experimetria applications scientist is recommended with the Working Heart system. Experimetria's systems are of the highest commercial quality available for complete whole heart perfusion research. Experimetria's systems are also completely modular and so they could be tailored to a specific researchers needs as requested. In addition to offering the complete perfusion apparatus, Experimetria also offers high quality transducers, amplifiers, probes and data acquisition/analysis electronics and software. Applications include testing inotropic, chronotropic, arrhythmogenic, anti-arrhythmic, anti-ischemic, vasoactive (coronary) effects in terms of functional, electrophysiological and biochemical parameters. |

The system is designed to measure the following parameters:
Working Heart cannula outside diameter (O.D.)
A smaller O.D. is not recommended for the working heart system because the pressure measurement is done with a catheter positioned into the left ventricular through the atrial cannula. With a smaller O.D. the atrial cannula would be closed by the pressure measuring catheter and the flow would be blocked. Thus the Working Heart system can not be used with mouse hearts. The given O.D. sizes are both for aortic cannula and atrial cannula. Langendorff cannula outside diameter (O.D.)

Experimetria does offer a mouse Langendorff. Here, the left ventricular measuring is done by a balloon which is fixed on a cannula that is attached to a pressure sensor. Cannulas smaller than 0.9mm are not recommended because the flow will drop down dramatically under this O.D. (with this O.D. the flow is ~60 ml per minute). For each cannula size desired, you will need to get a specific heart suspending unit. Special sizes of cannulas can be provided upon request for both systems however we can not exceed the extreme limits of size outlined here.
